Plant Transformation Vectors

Vera Thole & Philippe Vain

pCLEAN dual binary vector system

Added features of pCLEAN vectors:
(compared to previous pGreen and pSoup-based vectors)
  •  Counter selection of backbone transfer
  •  Modified left border (LB) of T-DNA
  •  Minimal T-DNA (only 52 nt excluding the borders)
  •  Reduced homology between pCLEAN and pSoup
  •  Multiple modified LBs
  •  Additional vir genes
  •  Exploiting backbone transfer for stable transformation
  •  Delivering multiple T-DNAs (clean-gene transformation)

pBrachyTAG dual binary vector system

Added features of pBrachyTAG vectors:
(compared to previous pGreen and pSoup-based vectors)

  • Modified left border (LB) of T-DNA

  • Reduced homology between pBrachyTAG and pSoup

  • Multiple modified LBs

  • Additional vir genes

  • UniBox sequence to facilitate the retrieval of FSTs

  • Promoter trapping features

The pCLEAN, pBrachyTAG and pGreen/pSoup dual binary vector systems are fully compatible enabling pGreen-based and pSoup-based vectors to be mixed and matched in Agrobacterium.
