Arabidopsis Libraries




BC Library

A binary cosmid library consisting of 73,728 clones, constructed using pCLD04541 as vector. This library has an average insert size of 17kb and was made at JIC using total DNA from the leaves of Arabidopsis thaliana, ecotype Columbia, partially digested with TaqI and ligated into the ClaI site of the vector

K Library

A TAC library consisting of 9,216 clones, constructed using pYLTAC7 as vector. This library has an average insert size of 80kb. JAtC Library

A large-insert binary vector library consisting of 36,096 clones, constructed using pCLD04541 as vector. This library provides 8x coverage of the Arabidopsis genome and was made at JIC using DNA extracted from purified nuclei from the leaves of Arabidopsis thaliana, ecotype Columbia, partially digested with Sau3AI and ligated into the BamHI site of the vector. This library utilizes Tetracycline selection in E. coli and Agrobacterium and selection of transformed plants is by resistance to Kanamycin.
This library is divided into the following sub-libraries:

JAtY Library

A TAC library consisting of 36,864 clones, constructed using pYLTAC17 as vector. This library provides 14x coverage of the Arabidopsis genome and was made at JIC using DNA extracted from purified nuclei from the leaves of Arabidopsis thaliana, ecotype Columbia, partially digested with HindIII and ligated into the HindIII site of the vector. This library utilizes Kanamycin selection in E. coli and Agrobacterium and selection of transformed plants is by resistance to phosphinothricin (Basta).
This library is divided into the following sub-libraries:
