M2 generation

 

Seed store

 

We decided to continue with the 0.30 and 0.40-treated plants, and sowed out ten seeds for each M1 line. These seeds were carefully counted out by Harriet Saunders, Alicia Grix and Pauline.

The ten seeds were sown in 1-liter pots by JIC horticultural staff during August 2007.

We used ten seeds so that most lines would produce at least two plants. After germination, seedlings would be removed to allow only two M2 plants per pot.

During autumn of 2007, leaf tissue was taken from each plant and placed in 96-well plates for DNA isolation by the JIC Genome lab (JGL).  The plants were then Individually bagged by JIC horticultural staff.

Finally, all the M2 plants were threshed during the summer of 2008 by Lucy, Harriet and Harry (picture) and Shirley Aris.

… and now both the M2 and M3 seeds are safely stored in the JIC seed storage facility.
